Abstract
Processes and systems related to solid phase peptide synthesis are described. The processes include a flow-through process comprising cyclic addition of amino acids to a column packed with resin, wherein each cycle includes the combination of the amino acids with one or more reagents to provide an activated amino acid mixture, and wherein heating is applied to the amino acid(s) before they are passed through the column and wherein the amino acids are re-circulated at least once over the column packed with resin. Systems for such processes are also described.
Claims12
A system for flow-through solid phase peptide synthesis (SPPS), wherein the system comprises:
- at least one column packed with resin;
- at least one pump;
- reservoirs for amino acids and reagents, wherein amino acid(s) are combined with one or more reagent to provide an amino acid mixture;
- at least one temperature regulating device arranged downstream of the reservoirs but upstream of the at least one column to heat the amino acid mixture;
- at least one temperature sensor;
- at least one detector arranged downstream of the at least one column to monitor progress of peptide synthesis in the at least one column;
- a re-circulation loop arranged to recirculate the amino acid mixture at least once over the at least one column;
- and
- at least one processor, wherein
- tubing connects the reservoirs to the column;
- valves are arranged to direct the flow in the system;
- and
- the processor is in communicative connection at least with the pump, the valves, the detector and the temperature regulating device.
