Abstract
Disclosed herein are methods of increasing nitrogen fixation in a non-leguminous plant. The methods can comprise exposing the plant to a plurality of bacteria. Each member of the plurality comprises one or more genetic variations introduced into one or more genes or non-coding polynucleotides of the bacteria's nitrogen fixation or assimilation genetic regulatory network, such that the bacteria are capable of fixing atmospheric nitrogen in the presence of exogenous nitrogen. The bacteria are not intergeneric microorganisms. Additionally, the bacteria, in planta, produce 1% or more of the fixed nitrogen in the plant.

An engineered diazotrophic bacterium comprising one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation, wherein:
- the one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation comprise a complete or partial deletion of at least one gene of the nitrogen fixation or assimilation genetic regulatory network, a heterologous promoter operably linked to at least one gene of the nitrogen fixation or assimilation genetic regulatory network, or a combination thereof;
- and
- the one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation are non-intergeneric.
An engineered diazotrophic bacterium comprising one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation, wherein:
- the one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation comprise:
- a complete or partial deletion of nifL, and
- a heterologous promoter operably linked to nifA;
- and
- the one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation are non-intergeneric.
An engineered diazotrophic bacterium comprising one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation, wherein:
- the one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation comprise:
- a complete or partial deletion of nifL,
- a heterologous promoter operably linked to nifA, and
- a complete or partial deletion of glnE, wherein the complete or partial deletion of glnE is a deletion of the nucleic acid sequence encoding the adenylyl-removing domain of GlnE;
- and
- the one or more genetic modifications in one or more genes for regulating nitrogen fixation or assimilation are non-intergeneric.
