Abstract
The present disclosure provides a system for mimicking the secondary lymphoid organs where suspension cells (e.g., T cells) are expanded; methods of expanding, activating, and transfecting the suspension cells in the synthetic microenvironment, and suspension cells produced by such systems and methods.

1-68. (canceled)
A method of expanding, activating, and transfecting T cells, the method comprising:
- introducing T cells from a blood sample of a patient to a bioreactor comprising a macroporous scaffold;
- activating the T cells;
- expanding the T cells;
- transfecting the T cells;
- preparing the T cells for transfusion into the patient;
- and
- transfusing the T cells into the patient;
- wherein the macroporous scaffold comprises (i) gelatin or other extracellular matrix components and (ii) one or more antibodies that are specific for activation of the T cells.
The method of 69, wherein the gelatin or other extracellular matrix components comprise collagen.
The method of 69, wherein the macroporous scaffold is functionalized with the one or more antibodies.
The method of 69, wherein the T cells are expanded in the presence of one or more cytokines.
A method of treating a cancer of a patient, the method comprising:
- introducing T cells from a blood sample of the patient to a bioreactor comprising a macroporous scaffold;
- activating the T cells;
- expanding the T cells;
- transfecting the T cells;
- preparing the T cells for transfusion into the patient;
- and
- transfusing the T cells into the patient;
- wherein the macroporous scaffold comprises (i) gelatin or other extracellular matrix components and (ii) one or more antibodies that are specific for activation of the T cells.
The method of 84, wherein the gelatin or other extracellular matrix components comprise collagen.
