Nlrp3 inflammasome inhibitors and uses thereof

A compound having the structure of Formula (Ia), or a pharmaceutically acceptable salt or a stereoisomer thereof:

• X is —S—;
• R1A is hydrogen or methyl;
• R1B is hydrogen or methyl;
• R3 is C3-C12cycloalkyl, or 4 to 12 membered heterocycloalkyl;
• each of which is optionally substituted with 1, 2, or 3 R8;
• R8 is each independently selected from halogen, —OH, —ORa, —SH, —SRa, SF5, —NRcRd, C1-C6alkyl, C1-C6haloalkyl, C1-C6hydroxyalkyl, C1-C6heteroalkyl, C1-C6aminoalkyl, and C3-C6cycloalkyl;
• RZN is hydrogen, C1-C6alkyl, or C1-C6haloalkyl;
• each R6 is independently halogen, —OH, C1-C6alkyl, C1-C6haloalkyl, or C1-C6hydroxyalkyl;
• or two R6 are taken together with the atoms to which they are attached to form a C4-C8 cycloalkyl or 5 to 6-membered heterocycloalkyl, each of which is optionally substituted with one or more R12;
• each R12 is independently halogen, —OH, —CN, —NO2, —ORa, —NRcRd, C1-C6alkyl, C1-C6alkoxy, C1-C6haloalkyl, C1-C6hydroxyalkyl, or C1-C6aminoalkyl;
• each Ra is independently C1-C6alkyl or C1-C6haloalkyl;
• each Rc and Rd are independently hydrogen, C1-C6alkyl or C1-C6haloalkyl;
• and
• p is 1, 2, 3, or 4.
