Abstract
The present application relates to a bi-functional therapeutic for treating cancer that includes a targeting component which targets a tumor-associated antigen and an enzyme which, when delivered to a tumor by said targeting component, converts the tumor phenotype to that of an incompatible allograft or xenograft. The enzyme is coupled to the targeting component. Also disclosed is a method for treating cancer comprising administering the bi-functional therapeutic.

- i) a targeting component which targets a tumor-associated antigen, wherein the targeting component comprises an antibody or antigen-binding fragment thereof;
- and
- ii) an enzyme, wherein the enzyme is a glycosyltransferase selected from:
- a glycosyltransferase A (Alpha 1-3-N-Acetylgalactosaminyltransferase) or an allelic variant thereof or an enzymatically active portion thereof;
- a glycosyltransferase B (alpha 1-3-galactosyltransferase) or an allelic variant thereof or an enzymatically active portion thereof;
- an alpha-gal-transferase or an allelic variant thereof or an enzymatically active portion thereof;
- a glycosyltransferase A (Gly268Ala) or an allelic variant thereof or an enzymatically active portion thereof;
- and
- a fucosyltransferase or an allelic variant thereof or an enzymatically active portion thereof.
