Abstract
The invention is directed to a bispecific chimeric antigen receptor, comprising: (a) at least two antigen-specific targeting regions; (b) an extracellular spacer domain; (c) a transmembrane domain; (d) at least one co-stimulatory domain; and (e) an intracellular signaling domain, wherein each antigen-specific targeting region comprises an antigen-specific single chain Fv (scFv) fragment, and binds a different antigen, and wherein the bispecific chimeric antigen receptor is co-expressed with a therapeutic control. The invention also provides methods and uses of the bispecific chimeric antigen receptors.

A bispecific chimeric antigen receptor (CAR), comprising, in N-terminal to C-terminal order:
- a) an anti-CD20 scFv comprising a VH and a VL, wherein the VH and VL amino acid sequences comprise the anti-CD20 VH and VL amino acid sequences, respectively, in SEQ ID NO:
- 3 or 12;
- b) a linker;
- c) an anti-CD19 scFv comprising a VH and a VL, wherein the VH and VL amino acid sequences comprise the anti-CD19 VH and VL amino acid sequences, respectively, in SEQ ID NO:
- 3 or 12;
- d) an IgG4 hinge extracellular spacer domain;
- e) a CD28 transmembrane domain;
- f) a 4-1BB co-stimulatory domain;
- and
- g) a CD3 zeta intracellular signaling domain.
