Abstract
There is provided a system for separation of analytes in a solution. The system encompasses a disposable cartridge enclosing a sorbent for binding the analytes in a solution and a conduit establishing a fluid link to a valve having a holding-loop to achieve elution through the cartridge at low pressures. The valve is switchable to a position following the elution from the cartridge for emptying through an outlet port at high pressures such as those required for moving liquid through chromatographic columns.
Claims29
A method for separation of analytes, said method comprising the steps of:
- placing a disposable cartridge enclosing a first sorbent binding said analytes in a recipient socket configured for receiving the cartridge, and for allowing a first fluid to flow through the cartridge into said recipient socket, thereby allowing for the elution of said analytes into said recipient socket;
- establishing a fluid link from said recipient socket to a valve by a conduit in fluid communication with said recipient socket and said valve, said valve being switchable between a first valve position and a second valve position, wherein when said valve is in said first valve position fluid communication is provided between said recipient socket and a holding-loop without sorbent, and wherein when said valve is in said second valve position fluid communication is provided between said holding-loop and an outlet port to provide for emptying said holding-loop through said outlet port;
- establishing a fluid link from said outlet port to a chromatographic column by a conduit in fluid communication with said outlet port and said chromatographic column, said chromatographic column being provided with a second sorbent;
- eluting said analytes from said disposable cartridge by providing flow of said first fluid through said disposable cartridge, said recipient socket, and said holding-loop by use of a first pump arrangement when said valve is in said first valve position;
- and
- providing flow of said first fluid through said holding-loop to said outlet port and said chromatographic column by use of a second pump arrangement when said valve is in said second valve position,
- wherein said holding-loop is configured to provide low flow resistance at a first flow rate of between 200 nL/min and 50 μL/min enabling elution of the analytes from said disposable cartridge and into said holding-loop at a first pressure of between 1 bar and 50 bar, and
- wherein said holding-loop is emptied through said outlet port at a second pressure of between 1 bar and 3000 bar and at a second flow rate of between 50 nL/min and 20 μL/min.
A method for separation of analytes, comprising:
- providing the analytes in a disposable cartridge having a first sorbent, the first sorbent being configured to bind the analytes;
- placing the disposable cartridge in a recipient socket that is in fluid communication with a valve, the valve being switchable between a first valve position and a second valve position, the first valve position placing the disposable cartridge in fluid communication with a holding-loop without sorbent, the second valve position placing the holding loop in fluid communication with a chromatographic column and a high-pressure pump, wherein the chromatographic column has a second sorbent;
- pumping a fluid through the disposable cartridge to the holding-loop with a low-pressure pump with the valve in the first valve position at a first flow rate of between 200 nL/min and 50 μL/min and a first pressure of between 1 bar and 50 bar so that the analytes in the disposable cartridge bind to the first sorbent;
- switching the valve from the first valve position to the second valve position;
- and
- pumping the fluid through the holding-loop to the chromatographic column with the high-pressure pump when the valve is in the second valve position at a second flow rate of between 50 nL/min and 20 μL/min and a second pressure of between 1 bar and 3000 bar.
