Abstract
Provided herein is a biological substance treatment method in which a flow cell is prepared, the flow cell including a chamber array device including a plurality of chambers and a cover body defining a flow path communicating with an opening of the plurality of chambers and provided in common for the plurality of chambers.
Claims17
A biological substance treatment method for retaining and/or enriching a biological substance, the biological substance treatment method comprising:
- a flow cell preparation step of preparing a flow cell comprising a chamber array device including a plurality of chambers each having a recess opened at one end, one of which is 1 nanoliter or less, and a cover defining a flow path communicating with the opening of the chamber and provided in common for the plurality of chambers;
- a solvent preparation step of preparing a first solvent and a second solvent that are aqueous to each other and separate into phases when left to stand at room temperature, the first solvent having a property of separating to the chamber side when phase separation occurs in a region surrounded by the chamber and the flow path of the flow cell, and the first solvent and the second solvent having a property that the biological substance is preferentially distributed to the first solvent rather than the second solvent;
- a mixing step of mixing the biological substance with the first solvent and/or the second solvent;
- a first inflow step of causing one solvent of the first solvent and the second solvent to flow into the flow path to fill the chamber and the flow path with the solvent;
- and
- a second inflow step of causing the first solvent, the other solvent of the first solvent and the second solvent, and the other of the second solvent to flow into the flow path to phase-separate the first solvent into the chamber side and the second solvent into the flow path side, retaining, in the chamber, the biological substance contained in the first solvent in the chamber without diffusing into the second solvent, and/or moving the biological substance contained in the second solvent into the first solvent in the chamber to enrich the biological substance.
A biological substance treatment apparatus for retaining and/or enriching a biological substance, the biological substance treatment apparatus comprising:
- a flow cell comprising a chamber array device comprising a plurality of chambers each having a recess opened at one end, one of which is 1 nanoliter or less, and a flow path communicating with the opening of the chamber and provided in common for the plurality of chambers;
- a first solvent and a second solvent that are aqueous to each other and separate into phases when left to stand at room temperature, the first solvent having a property of separating to the chamber side when phase separation occurs in a region surrounded by the chamber and the flow path of the flow cell, and the first solvent and the second solvent having a property that the biological substance is preferentially distributed to the first solvent rather than the second solvent;
- means for mixing the biological substance with the first solvent and/or the second solvent;
- first inflow means for causing one solvent of the first solvent and the second solvent to flow into the flow path to fill the chamber and the flow path with the solvent;
- and
- second inflow means for causing the other solvent of the first solvent and the second solvent to flow into the flow path to phase-separate the first solvent into the chamber side and the second solvent into the flow path side, bringing the first solvent and the second solvent into contact with each other, retaining, in the chamber, the biological substance contained in the first solvent in the chamber without diffusing into the second solvent, and/or moving the biological substance contained in the second solvent into the first solvent in the chamber to enrich the biological substance.
A biological substance treatment apparatus for retaining and/or enriching abiological substance using a flow cell comprising a chamber array device comprising a plurality of chambers each having a recess opened at one end, one of which is 1 nanoliter or less, and a flow path communicating with the opening of the chamber and provided in common for the plurality of chambers, the biological substance treatment apparatus comprising:
- a first solvent and a second solvent that are aqueous to each other and separate into phases when left to stand at room temperature, the first solvent having a property of separating to the chamber side when phase separation occurs in a region surrounded by the chamber and the flow path of the flow cell, and the first solvent and the second solvent having a property that the biological substance is preferentially distributed to the first solvent rather than the second solvent;
- means for mixing the biological substance with the first solvent and/or the second solvent;
- first inflow means for causing one of the first solvent and the second solvent to flow into the flow path to fill the chamber and the flow path with the solvent;
- and
- second inflow means for causing the other of the first solvent and the second solvent to flow into the flow path to phase-separate the first solvent into the chamber side and the second solvent into the flow path side, retaining, in the chamber, the biological substance contained in the first solvent in the chamber without diffusing into the second solvent, and/or moving the biological substance contained in the second solvent into the first solvent in the chamber to enrich the biological substance.
