Abstract
Provided herein are compositions for a Herpes Simplex Virus packaging system and packaging cell line. Also provided herein are compositions, systems, and methods for producing a packaging cell line for Herpes Simplex Virus amplicons.
Claims157
A packaging cell line comprising a recombinant genome, wherein the recombinant genome comprises:
- a. a first nucleic acid sequence comprising one or more Herpes Simplex Virus (HSV) Class I genes;
- and
- b. a second nucleic acid sequence comprising one or more HSV Class I and Class II genes.
The kit of claim of 61, wherein the amplicon vector further comprises at least one cargo sequence.
The kit of claim of 62, wherein the at least one cargo sequence comprises a transgene selected from the group consisting of CRISPR associated protein 9 (Cas9), Cas3, Cas12a, Dystrophin (DYS), amylo-alpha-1, 6-glucosidase, 4-alpha-glucanotransferase (AGL), otoferlin (OTOF), myosin VIIA (MYO7A), ATP binding cassette subfamily A member 4 (ABCA4), coagulation factor VIII (F8), centrosomal protein 290 (CEP290), cadherin related 23 (CDH23), or centrosome and basal body associated protein (ALMS1).
The kit of any one of claims of 62-64, wherein the at least one cargo nucleic acid sequence further comprises at least one regulatory element comprising an enhancer, a promoter, a terminator, or a combination thereof.
A composition comprising a recombinant genome, wherein the recombinant genome comprises:
- a. a first nucleic acid sequence comprising one or more Herpes Simplex Virus (HSV) Class I genes;
- and
- b. a second nucleic acid sequence comprising one or more HSV Class I and Class 11 genes.
A method of producing a packaging cell line comprising introducing into a cell comprising a genome:
- a. a first nucleic acid sequence comprising one or more Herpes Simplex Virus (HSV) Class I genes;
- and
- b. a second nucleic acid sequence comprising one or more HSV Class I and Class II genes.
