Abstract
The present disclosure relates to protein production in mammalian cell lines using artificial neural network-like genetic circuits, and convolutional neural network predictions are described. A genetic circuit system including gene nodes selected based on roles in view of a target biological output is provided. The genetic circuit is applied to mammalian cells under bioproduction conditions, and the yield of the produced protein is measured.
Claims16
A genetic circuit system comprising:
- a data processing module configured to receive gene expression data for a plurality of genes from a plurality of biological samples;
- a feature selection module coupled to the data processing module and configured to identify a subset of key genes from the plurality of genes based on a target biological output;
- and
- a neural network model stored in a memory component and comprising a plurality of nodes arranged in a plurality of layers, wherein each node of the plurality of nodes corresponds to a gene of the subset of key genes and is associated with a gene-specific transfer function.
An engineered CHO cell, comprising an overexpressed sdhaf3 gene, an overexpressed psat1 gene, and an overexpressed fam83d gene.
