Abstract
The invention relates to a DNA origami biosensor capable of detecting at least two different target oligonucleotides at the same time, said biosensor comprising at least one DNA origami hinge and at least two detection sites, wherein each detection site comprises a signal emitting array, at least one lock and at least one movable section, wherein the movable sections of the detection sites are connected to each other or to a common DNA origami structure via the at least one hinge.
Claims16
A DNA origami biosensor capable of detecting at least two different target oligonucleotides at the same time, said biosensor comprising at least one DNA origami hinge and at least two detection sites, wherein each detection site comprises
- a signal emitting array,
- at least one lock comprising a first and a second fastening domain and a toehold domain attached to the first or second fastening domain, wherein the first and the second fastening domain are capable of forming a duplex with each other and wherein the first or second fastening domain is capable of hybridizing to a target oligonucleotide via the toehold domain, wherein the lock is capable of assuming an open or a locked configuration, wherein the lock assumes the open configuration upon hybridization of the target oligonucleotide and the locked configuration in the absence of the target oligonucleotide,
- at least one movable section, wherein the movable section is connected to the at least one hinge and is movable via said hinge and wherein the position of the movable section depends on the configuration of the lock,
- wherein the movable sections of the detection sites are connected to each other or to a common DNA origami structure via the at least one hinge.
Dependencies are inferred from the claim text.
