A method, comprising:
- obtaining or receiving a fluid sample from a subject via a sample container of a device, wherein the fluid sample comprises a plurality of fragmented cell-free DNA molecules comprising one or more methylation markers;
- amplifying the plurality of fragmented cell-free DNA molecules with an amplification reagent on the device thereby producing a plurality of amplified fragments of cell-free DNA molecules, wherein the amplification reagent comprises multiple primer pairs;
- and
- determining a disease of the subject from the one or more methylation markers of the plurality of amplified fragments of cell-free DNA molecules.

A system, comprising:
- a device configured to collect or obtain a fluid sample from a subject via a sample container of the device, wherein the fluid sample comprises a plurality of fragmented cell-free DNA molecules comprising one or more methylation markers,
- wherein the device comprises a processor configured to perform executable instructions, and
- wherein the device comprises a program comprising instructions executable by the processor to:
- amplify the plurality of fragmented cell-free DNA molecules with an amplification reagent on the device thereby producing a plurality of amplified fragments of cell-free DNA molecules, wherein the amplification reagent comprises multiple primer pairs:
- obtain a plurality of signals corresponding to the plurality of amplified fragments of cell-free DNA molecules with a detector on the device;
- and
- determine a disease of the subject from the plurality of signals based at least on the one or more methylation markers of the plurality of amplified fragments of cell-free DNA molecules.
