A delivery vector including a DNA sequence encoding pre-prodynorphin or pre-prodynorphin-variants, which drives expression of a pre-propeptide in a target cell, and enables the release of dynorphin or dynorphin-variants from the target cell on demand, and contains a signal peptide, which is a N-terminal extension of nascent polypeptide chains and mediates protein targeting to the lumen of the endoplasmatic reticulum, and contains a N-terminal pro-peptide fragment on the C-terminal end and contains i) a sorting motif including the elements DL and EXyL, or ii) a sorting motif of a pre-pro-neuropeptide or protein sorted to large dense core vesicles, other than pre-prodynorphin, where in certain embodiments x is an integer 1 to 20, y is an integer 1 to 10, and each X is independently an amino acid, and wherein the N-terminal pro-peptide fragment consists of 16 to 90 amino acids, and corresponding peptides, DNA molecules viral vectors and particles.
