Abstract
The present disclosure relates to cellular therapy products comprising mesenchymal lineage precursor or stem cells and potency assay for these products. The present disclosure also relates to methods for treatment of immune or inflammatory disorders, and treatment or prevention of graft versus host disease (GVHD), or one or more symptoms associated with GVHD, by administration of mesenchymal lineage precursor or stem cells.
Claims13
A method for selecting a population of mesenchymal lineage or stem cells (MLPSCs) suitable for use in therapy comprising:
- (i) obtaining a population of MLPSCs that has been culture expanded,
- (ii) co-culturing the obtained population of MLPSCs in a culture medium with a population of cells comprising T cells, wherein the population of cells comprising T cells is a population of CD3/CD28-activated peripheral blood mononuclear cells (PBMCs) and, wherein the co-culture comprises MLPSCs and activated PBMCs at a ratio of 1 MLPSC:5 PMBC or less;
- (iii) selecting a population of MLPSCs which inhibit T cell IL-2Rα expression by ≥65%.
A cryopreserved composition comprising a population of mesenchymal lineage or stem cells (MLPSCs), Plasmalyte-A, 25% human serum albumin (HSA) and DMSO, wherein the mesenchymal lineage precursor or stem cells are culture expanded mesenchymal stem cells;
- wherein the population inhibits expression of IL-2Rα in a sample of peripheral blood mononuclear cells (PBMCs) comprising activated T-cells by at least 65%;
- and
- wherein the inhibition of expression of IL-2Rα is measured by co-incubation of the population and the sample of PBMCs comprising activated T cells at a ratio of 1 MSC:5 PBMC or less, wherein the level of inhibition of IL-2Rα expression by the MSCs is determined in the activated T cells.
