Abstract
The present disclosure provides engineered mesenchymal stem cells (MSCs), a pharmaceutical composition containing the same, and use thereof in the manufacture of a medicament for treating cancer. The mesenchymal stem cells (MSCs) comprise an introduced NQO1 protein-coding sequence and/or an introduced hypoxia response element (HRE), and a nucleic acid encoding one or more immunostimulatory cytokines.
Claims21
1-25. (canceled)
A method for treating cancer, comprising:
- administering a therapeutically effective amount of engineered mesenchymal stem cells (MSCs) to a subject in need thereof,
- wherein the engineered mesenchymal stem cell includes in its genome an introduced NQO1 protein-coding sequence, an introduced regulatory element, and an introduced target gene,
- wherein the expression of the target gene is regulated by the regulatory element;
- wherein the target gene encodes an anti-tumor protein, preferably an immunostimulatory cytokine or an anti-tumor cytokine.
An engineered mesenchymal stem cell (MSC) comprising in its genome an introduced NQO1 protein-coding sequence, an introduced regulatory element, and an introduced target gene,
- wherein the expression of the target gene is regulated by the regulatory element;
- wherein the target gene encodes an anti-tumor protein, preferably an immunostimulatory cytokine or an anti-tumor cytokine.
A method for delivering a target gene to tumor tissue, including using an engineered mesenchymal stem cell (MSC) as a carrier of the target gene, wherein the MSC includes in its genome an introduced NQO1 protein-coding sequence and the target gene.
