Abstract
Provided are methods for determining the amount of tamoxifen and its metabolites in a sample by mass spectrometry. In some aspects, the methods provided herein determine the amount of N-Desmethyl Tamoxifen. In some aspects, the methods provided herein determine the amount of N-Desmethyl Tamoxifen and other tamoxifen metabolites. In some aspects, the methods provided herein determine the amount of tamoxifen, N-Desmethyl Tamoxifen, and other tamoxifen metabolites.
Claims8
A method for determining an amount of tamoxifen and at least one tamoxifen metabolite in a sample in a single mass spectrometry assay, comprising:
- purifying a sample by liquid chromatography;
- ionizing tamoxifen and at least one tamoxifen metabolite to produce one or more ions detectable by mass spectrometry, wherein the at least one tamoxifen metabolite is selected from the group consisting of norendoxifen, endoxifen, 4′-Hydroxy Tamoxifen, 4-Hydroxy Tamoxifen, N-Desmethyl-4′-Hydroxy Tamoxifen, and N-Desmethyl Tamoxifen;
- and
- detecting an amount of the ion(s) from step (b) by mass spectrometry, wherein the amount of the ion(s) detected is related to the amount of each of tamoxifen and the at least one tamoxifen metabolite in the sample,
- wherein the method has a limit of quantitation of less than or equal to 1.5 ng/mL for tamoxifen.
