Abstract
Provided is a parent phytase variant, which relates to the technical field of protein engineering. The variant, relative to the parent phytase thereof, has one or more amino acid substitutions at positions corresponding to positions 295, 349, and 374 of SEQ ID NO: 1. Compared to the parent phytase, the variant has increased thermal stability.

A thermostable phytase variant, which
- (a) has one or more amino acid substitutions at positions corresponding to positions 295, 349, and 374 of the polypeptide of SEQ ID NO:
- 1, wherein the amino acid position corresponding to position 295 of the polypeptide of SEQ ID NO:
- 1 is substituted with P, Y, G, K, L or Q, or wherein the amino acid position corresponding to position 349 of the polypeptide of SEQ ID NO:1 is substituted with K, L, G, H, R, or T;
- (b) has at least 90% sequence identity to the polypeptide of SEQ ID NO:
- 98;
- and
- (c) has phytase activity.
