Abstract
The present invention relates to the use of tissue non-specific alkaline phosphatase (TNAP) as a marker for identifying and/or isolating adult multipotential cells. The present invention also relates to cell populations enriched by methods of the present invention and therapeutic uses of these cells.

1-45. (canceled)
A method of enriching a population of cells for adult multipotential cells, the method comprising;
- (i) contacting a sample which comprises the population of adult multipotential cells with an anti-TNAP antibody under conditions that allows binding of the anti-TNAP antibody to cells expressing TNAP and obtaining a population of cells bound to the anti-TNAP antibody;
- and
- (ii) culturing the population of cells obtained in step (i) to enrich for multipotential cells expressing the STRO-1 marker, wherein the anti-TNAP antibody is an antibody that binds to the same epitope as the STRO-3 antibody produced by the hybridoma cell line deposited with the American Type Culture Collection on 19 Dec. 2005 pursuant to the provisions of the Budapest Treaty, under Accession Number PTA-7282.
A method of generating a population of tissue specific, committed cells, the method comprising;
- (i) contacting a sample which comprises a population of adult multipotential cells with an anti-TNAP antibody under conditions that allow binding of TNAP to the anti-TNAP antibody and obtaining a population of cells bound to the anti-TNAP antibody;
- (ii) culturing the population of cells obtained in step (i) to enrich for multipotential cells expressing the STRO-1 marker;
- (iii) culturing the population of cells from step (ii) in the presence of one or more stimulatory factors;
- and
- (iv) subjecting the cultured population of cells to conditions biasing differentiation of adult multipotential cells to a specific tissue type so as to generate the population of tissue specific, committed cells.
