Abstract
The present invention relates to zebrafish embryo and adult models constructed by inducing inflammation with carboxymethyllysine (CML), which causes glycation toxicity, and a method for screening anti-inflammatory agents using the same. The present invention provides a method for screening a novel anti-inflammatory agent using zebrafish in which embryonic death, acute cytokine storm, and neuro-paralysis has been induced, whereby inflammatory diseases can be prevented or treated in a more convenient and cost-effective manner.
Claims19
A method for screening an anti-inflammatory agent comprising the following steps:
- 1) a step of inducing hyperinflammation, acute embryotoxicity, and acute cytokine storm by injecting CML into zebrafish embryos 15 to 45 minutes after fertilization;
- 2) a step of injecting CML together with the test substance into zebrafish embryos 15 to 45 minutes after fertilization;
- 3) a step of comparing the survival rate of the zebrafish embryos of step 2) and that of the zebrafish embryos of step 1);
- and
- 4) a step of determining that the zebrafish embryos of step 2) above exhibit normal development if they are at the primordium-3 to primordium-8 stage, have more than 30 somites, have distinct black eye pigmentation, and show a clear midbrain-hindbrain boundary (MHB) in the brain.
