Method and system for detecting one or more alleles of interest in a sample. In one embodiment, the method includes providing a multi-welled plate, each well having an electrochemical sensing element and a capture probe. The capture probe includes two or more nucleotide capture sequences, each designed to hybridize specifically with an allele of interest. The capture probe is exposed to a biofluid sample so that any alleles of interest present bind to the capture probe. Next, an initiator that is specific to only one of the alleles of interest is added to the well. If the allele of interest specific to the initiator is bound to the capture probe, the initiator binds to the allele of interest. Next, hairpin molecules that undergo hybridization chain reaction when the initiator is present are added. The hairpin molecules are coupled to a tag. The presence of the tag is then detected.
