Provided are nanoscale reaction chambers and methods of using the nanoscale reaction chambers for biological assays.

A method of detecting an interaction between a first cell and a second cell, the method comprising:
- (a) providing or obtaining a nanovial comprising a cavity formed therein and an affinity agent immobilized thereto;
- (b) loading a first cell into the cavity of the nanovial;
- (c) loading a second cell into the cavity of the nanovial, the second cell capable of interacting with the first cell;
- (d) generating at least one marker or readout produced by either the first cell or the second cell upon interaction between the first cell and the second cell;
- and
- (e) detecting the at least one marker or readout directly or with a detection agent to obtain a signal indicative of functional activity caused by the interaction between the first cell and the second cell.
