Abstract
The invention provides methods for genetically engineering T cells in vivo comprising administering to the subject a cytokine or nucleic acid molecule encoding a cytokine to recruit the subject's T cells to the administration site; followed by administration of a nucleic acid molecule encoding an antigen receptor. In some instances, the method also includes administering an integrase or nucleic acid molecule encoding an integrase to integrate the sequence encoding the antigen receptor into the DNA of the recruited T cells.
Claims26
A method for treating cancer in a subject in need thereof, comprising:
- (a) administering a cytokine composition at a treatment site of the subject thereby recruiting at least one T cell to the treatment site;
- and
- (b) administering an antigen receptor composition to the subject to genetically modify the recruited at least one T cell to express an antigen receptor.

A method for generating a tumor-specific T cell in a subject, comprising:
- (a) administering a cytokine composition at a treatment site of the subject thereby recruiting at least one T cell to the treatment site;
- and
- (b) administering an antigen receptor composition to the subject to genetically modify the recruited at least one T cell to express an antigen receptor that binds to a tumor-specific antigen.
