Abstract
The present invention provides nuclease-resistant nucleic acid nanostructures, pharmaceutical compositions thereof, pharmaceutical and diagnostic uses thereof as well as a method of producing nucleic acid nanostructures.
Claims20
A nucleic acid nanostructure, comprising:
- a) a core nucleic acid construct comprising a self-assembling nucleic acid building block;
- b) a coat oligonucleotide, wherein the coat oligonucleotide comprises a nuclease-resistant nucleotide, wherein the coat oligonucleotide is linked to at least a portion of the core nucleic acid construct;
- and
- wherein the nuclease-resistant nucleotide is located at pre-defined sites or at a density sufficient to allow the nucleic acid nanostructure to decompose in a time-controlled manner in the presence of a nuclease.

A method of producing a nucleic acid nanostructure, the method comprising:
- (a) providing a self-assembling nucleic acid building block that comprises a coat-binding oligonucleotide located at pre-defined sites or at a density sufficient to allow the nucleic acid nanostructure to decompose in a time-controlled manner in the presence of a nuclease;
- (b) providing a coat oligonucleotide comprising a nuclease-resistant nucleotide that comprises a nucleic acid sequence binds to a DNA sequence of the coat-binding oligonucleotide;
- and
- (b) subjecting the self-assembling nucleic acid building block and the coat oligonucleotide to a condition that allows for complementary base pairing, thereby producing the nucleic acid nanostructure.
