Abstract
RNA nanostructures with significantly reduced immunogenicity are described herein, enhancing their therapeutic utility. Incorporating specific base modifications during in vitro transcription decreases recognition by innate immune sensors. This breakthrough enables safer and more effective applications in targeted drug delivery and gene therapy, representing a significant advancement over current RNA-based technologies.
Claims20
An RNA nanostructure comprising a single RNA strand that includes a plurality of modified nucleotides, wherein the RNA strand is configured to self-assemble into the RNA nanostructure.
A method of reducing innate immunogenicity in a cell, the method comprising contacting the cell with an RNA nanostructure comprising a single RNA strand that includes a plurality of modified nucleotides, wherein the RNA strand is configured to self-assemble into the RNA nanostructure.
A method of reducing innate immunogenicity in a subject in need thereof, the method comprising:
- administering an RNA nanostructure comprising a single RNA strand that includes a plurality of modified nucleotides, wherein the RNA strand is configured to self-assemble into the RNA nanostructure.
