Mrgprx2 modulators and related methods of treatment

A compound having the structure of Formula (III-A):

• or a pharmaceutically acceptable isomer, racemate, hydrate, solvate, isotope, or salt thereof, wherein:
• Q1 is N or CR6, and Q2 and Q3 are CR6;
• R3 is H or C1-6 alkyl;
• R4 is, at each occurrence, independently halo, —CN, —OR7, —N(R7)2, C1-6 alkyl, C1-6 haloalkyl;
• R5 is, at each occurrence, independently halo, —CN, —OR7, —N(R7)2, —S(O)2—R7, C1-6 alkyl, C1-6 haloalkyl, carbocycle, or heterocycle;
• R6 is, at each occurrence, independently halo, —CN, —OR7, —N(R7)2, —SR7, —C(O)OR7, —N(R8)C(O)R7, —N(R8)C(O)OR7, —S(O)2—R7, —C1-6 alkyl-R7, C1-6 haloalkyl, carbocycle substituted with —(R7)x, or heterocycle substituted with —(R7)x;
• R7 is, at each occurrence, independently H, —OR8, —N(R8)2, —C(O)OR8, —NRC(O)OR8, —C1-6 alkyl-R8, C1-6 haloalkyl, carbocycle substituted with —(R8)y, or heterocycle substituted with —(R′)y;
• R8 is, at each occurrence, independently H, —OR, —NR2, —C1-6 alkyl-R, carbocycle substituted with —(R)z, or heterocycle substituted with —(R)z;
• R′ and R″ are, at each occurrence, independently H, halo, cyano, hydroxy, amino, C1-6 alkyl, or C1-6 alkenyl, or, independently, at each occurrence of R′ and R″ attached to the same carbon atom, R′ and R″, together with the carbon to which they are attached, form a cycloalkyl;
• Ra and Rb are, at each occurrence, independently H, halo, cyano, hydroxy, amino, or C1-6 alkyl or, independently, at each occurrence of Ra and Rb attached to the same carbon atom, Ra and Rb, together with the carbon to which they are attached, form a cycloalkyl;
• R is, at each occurrence, independently H, halo, —CN, —OH, —NH2, or C1-6 alkyl,
• m is 0-5;
• n is 0-5;
• p is 0-5
• x is 0-3;
• y is 0-3;
• and
• z is 0-3.
