Abstract
Methods are described for measuring the amount of a vitamin B2 in a sample. More specifically, mass spectrometric methods are described for detecting and quantifying vitamin B2 in a sample utilizing on-line extraction methods coupled with tandem mass spectrometric techniques.
Claims9
A method for determining the amount of vitamin B2 in a biological sample from a human, the method comprising:
- adding 13C, 15N2-vitamin B2 to the sample;
- purifying the sample by high performance liquid chromatography (HPLC);
- ionizing the 13C, 15N2-vitamin B2 and the vitamin B2 to produce a fragment ion of 13C, 15N2-vitamin B2 detectable by tandem mass spectrometry and a fragment ion of vitamin B2 detectable by tandem mass spectrometry;
- determining by mass spectrometry the amount of the fragment ion of the 13C, 15N2-vitamin B2 and the amount of the fragment of the vitamin B2;
- and
- comparing the amount of the fragment ion of vitamin B2 and the fragment ion of the 13C, 15N2-vitamin B2 to determine the amount of vitamin B2 in the sample;
- and
- wherein the method has a lower limit of quantitation of 5 nmol/L to 25 nmol/L, inclusive.
