## Abstract

A system for buoyant-particle-assisted cell therapy includes and/or interfaces with a set of buoyant particles. Additionally or alternatively, the system can include and/or interface with a processing container, a set of processing materials (e.g., buffers, factors, solutions, etc.), and/or any other components. A method for buoyant-particle-assisted cell therapy includes processing the set of cells of interest. Additionally or alternatively, the method can include any or all of: preparing a set of buoyant particles; receiving a sample; and isolating a set of cells of interest from the sample.

## Claims

### Claim 1

A method for buoyancy-assisted production of cells for a cell therapy, the method comprising:
producing bound T cells by combining a set of antibodies, a set of buoyant particles, and T cells within a container, wherein the set of buoyant particles are functionalized with moieties for binding to and forming a complex with the antibodies, wherein the bound T cells aggregate at a superior region of the container;
culturing the bound T cells in the container to produce daughter cells from the bound T cells, wherein the daughter cells detach from the bound T cells and aggregate at an inferior surface of the container, thereby decreasing exhaustion of the daughter cells due to exposure of the daughter cells to the antibodies; and
collecting the daughter cells, wherein the daughter cells are used in the cell therapy.

### Claim 2 (depends on 1)

The method of claim 1, wherein in at least a portion of the bound T cells, a single T cell is bound to multiple buoyant particles of the set of buoyant particles.

### Claim 3 (depends on 1)

The method of claim 1, wherein the bound T cells are cultured for at least 3 days.

### Claim 4 (depends on 1)

The method of claim 1, further comprising performing a selection process by combining the daughter cells with a second set of buoyant particles.

### Claim 5 (depends on 1)

The method of claim 1, wherein the daughter cells sink from the superior region of the container to the inferior surface of the container, thereby increasing a spatial separation between the daughter cells and the bound T cells.

### Claim 6 (depends on 1)

The method of claim 1, wherein the set of buoyant particles comprises glass beads.

### Claim 7 (depends on 1)

The method of claim 1, wherein the set of buoyant particles comprises streptavidin-functionalized buoyant particles.

### Claim 8 (depends on 1)

The method of claim 1, wherein a bound T cell in the bound T cells comprises multiple antibodies bound to a buoyant particle of the set of buoyant particles.

### Claim 9 (depends on 8)

The method of claim 8, wherein the multiple antibodies comprise a CD3 antibody and a CD28 antibody.

### Claim 10 (depends on 1)

The method of claim 1, wherein the set of buoyant particles are submerged within fluid in the container.

### Claim 11 (depends on 1)

The method of claim 1, wherein the container comprises a Leukopak bag.

### Claim 12 (depends on 1)

The method of claim 1, further comprising transfecting the T cells.

### Claim 13 (depends on 1)

The method of claim 1, wherein the container comprises a sealing mechanism configured to separate the superior region of the container from an inferior region of the container.

### Claim 14

A method for buoyancy-assisted production of cells for a cell therapy, the method comprising:
performing a first negative selection, comprising:
producing bound T cells by combining a first set of buoyant particles and T cells within a container, wherein the T cells comprise a first subset of the T cells and a second subset of the T cells, wherein the first set of buoyant particles are functionalized with moieties for binding to and forming a complex with the second subset of the T cells, wherein the bound T cells aggregate at a superior region of the container; and
removing the bound T cells;
culturing the first subset of the T cells in the container to produce expanded T cells;
performing a second negative selection, comprising:
producing second bound T cells by adding a second set of buoyant particles to the container, wherein the expanded T cells comprises a first subset of the expanded T cells and a second subset of the expanded T cells, wherein the second set of buoyant particles are functionalized with moieties for binding to and forming a complex with the second subset of the expanded T cells, wherein the second bound T cells aggregate at a superior region of the container; and
removing the second bound T cells; and
collecting the first subset of the expanded T cells for the cell therapy.

### Claim 15 (depends on 14)

The method of claim 14, wherein the first subset of T cells comprises CD8+ T cells.

### Claim 16 (depends on 14)

The method of claim 14, wherein the first subset of T cells CD4+ T cells.

### Claim 17 (depends on 14)

The method of claim 14, wherein in at least a portion of the bound T cells, a single T cell is bound to multiple buoyant particles of the first set of buoyant particles.

### Claim 18 (depends on 14)

The method of claim 14, wherein the first subset of the T cells is cultured for at least 3 days.

### Claim 19 (depends on 14)

The method of claim 14, wherein the container comprises a sealing mechanism configured to separate the superior region of the container from an inferior region of the container.

### Claim 20 (depends on 18)

The method of claim 18, wherein the sealing mechanism comprises a valve.
