Abstract
The present disclosure relates to anti-glyco-MUC1 antibodies and antigen binding fragments thereof that specifically bind to a cancer-specific glycosylation variant of MUC1 and related fusion proteins and antibody-drug conjugates, as well as nucleic acids encoding such biomolecules. The present disclosure further relates to use of the antibodies, antigen-binding fragments, fusion proteins, antibody-drug conjugates and nucleic acids for cancer therapy.

An anti-glyco-MUC1 antibody or antigen-binding fragment comprising:
- (a) a heavy chain variable (VH) sequence comprising a complementarity determining region (CDR) H1 comprising the amino acid sequence of SEQ ID NO:5, a CDR-H2 comprising the amino acid sequence of SEQ ID NO:6, and a CDR-H3 comprising the amino acid sequence of SEQ ID NO:7, and a light chain variable (VL) sequence comprising a CDR-L1 comprising the amino acid sequence of SEQ ID NO:8, a CDR-L2 comprising the amino acid sequence WVS, and a CDR-L3 comprising the amino acid sequence of SEQ ID NO:10;
- (b) a VH sequence comprising a CDR H1 comprising the amino acid sequence of SEQ ID NO:23, a CDR-H2 comprising the amino acid sequence of SEQ ID NO:24, and a CDR-H3 comprising the amino acid sequence of SEQ ID NO:25, and a VL sequence comprising a CDR-L1 comprising the amino acid sequence of SEQ ID NO:26, a CDR-L2 comprising the amino acid sequence of SEQ ID NO:27, and a CDR-L3 comprising the amino acid sequence of SEQ ID NO:10;
- or
- (c) a VH sequence comprising a CDR H1 comprising the amino acid sequence of SEQ ID NO:28, a CDR-H2 comprising the amino acid sequence of SEQ ID NO:29, a CDR-H3 comprising the amino acid sequence of SEQ ID NO:25, and a VL sequence comprising a CDR-L1 comprising the amino acid sequence of SEQ ID NO:30, a CDR-L2 comprising the amino acid sequence WVS, and a CDR-L3 comprising the amino acid sequence of SEQ ID NO:31.
