Abstract
Tissue explants of the gastrointestinal tract are provided. Methods of making and using the tissue explants are also provided, along with substrates designed for the tissue explants described.
Claims30
A high-throughput screening composition comprising:
- (i) a first plate comprising a plurality of microwells;
- (ii) a second plate comprising a plurality of microwells, wherein the number of microwells of the second plate matches the number of microwells of the first plate;
- (iii) an isolated tissue explant from a large, non-human, mammalian gastrointestinal tract placed in between the first plate and the second plate, wherein the tissue explant comprises epithelial cells having a polarity,
- wherein the plurality of microwells of the first plate are through holes, and wherein the plurality of microwells of the second plate are receiver chambers;
- and
- wherein the tissue explant is in planar contact with more than one microwell of the first plate and more than one microwell of the second plate, thereby providing a luminal surface and a basolateral surface of the tissue explant.
