Abstract
The present disclosure provides non-natural reverse transcriptases for conducting reverse transcription. The non-natural reverse transcriptases herein may have increased thermostability and can conduct reverse transcription more efficiently than natural reverse transcriptases.

A non-natural enzyme comprising a first domain and a second domain wherein:
- (a) the first domain is a non-ribonuclease enzyme domain;
- and
- (b) the second domain is a ribonuclease domain comprising a sequence having at least 80% sequence identity to SEQ ID NO:
- 7 or 8, wherein the first domain and the second domain are not derived from a same organism.
