The present disclosure relates to compositions of matter, methods, and systems for analyzing polymeric macromolecules, including polymeric macromolecules such as peptides, polypeptides, and proteins.

A method for sequencing a subset of nucleotides of an oligonucleotide, comprising:
- providing, in a nucleic acid sequencing reaction, a combination of reversibly terminated nucleotides and nucleotides that are not reversibly terminated;
- and
- performing the nucleic acid sequencing reaction, wherein nucleotides of the oligonucleotide being sequenced that correspond with the nucleotides that are reversibly terminated are sequenced, and wherein nucleotides of the oligonucleotide being sequenced that correspond with the nucleotides that are not reversibly terminated are not sequenced.
