Abstract
B-cell maturation antigen (BCMA) is expressed on malignant plasma cells. The present invention provides methods for treating multiple myeloma using bispecific antibodies (bsAbs) that bind to both BCMA and CD3 and activate T cells via the CD3 complex in the presence of BCMA-expressing tumor cells. In certain embodiments, the bispecific antigen-binding molecules of the present invention are capable of inhibiting the growth of tumors expressing BCMA.

A method of treating refractory multiple myeloma in a subject in need thereof, the method comprising administering a bispecific antibody comprising a first antigen-binding domain that specifically binds a human B-cell maturation antigen (BCMA), and a second antigen-binding domain that specifically binds human CD3, wherein:
- the first antigen-binding domain comprises:
- (a) a heavy chain variable region (HCVR) comprising three heavy chain complementarity determining regions, HCDR1, HCDR2 and HCDR3, wherein HCDR1, HCDR2 and HCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 68, 70 and 72, respectively;
- and
- (b) a light chain variable region (LCVR) comprising three light chain complementarity determining regions, LCDR1, LCDR2 and LCDR3, wherein LCDR1, LCDR2 and LCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 84, 86 and 88, respectively;
- and
- the second antigen-binding domain comprises:
- (a) a HCVR comprising three heavy chain complementarity determining regions, HCDR1, HCDR2 and HCDR3, wherein HCDR1, HCDR1 and HCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 92, 94 and 96, respectively;
- and
- (b) a LCVR comprising three light chain complementarity determining regions, LCDR1, LCDR2 and LCDR3, wherein LCDR1, LCDR2 and LCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 84, 86 and 88, respectively, and
- wherein the refractory multiple myeloma is triple refractory to an anti-CD38 antibody, a proteasome inhibitor, and an immunomodulatory drug.

- (a) a heavy chain variable region (HCVR) comprising three heavy chain complementarity determining regions, HCDR1, HCDR2 and HCDR3, wherein HCDR1, HCDR2 and HCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 68, 70 and 72, respectively;
- and
- (b) a light chain variable region (LCVR) comprising three light chain complementarity determining regions, LCDR1, LCDR2 and LCDR3, wherein LCDR1, LCDR2 and LCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 84, 86 and 88, respectively;
- and
- the second antigen-binding domain comprises:
- (a) a HCVR comprising three heavy chain complementarity determining regions, HCDR1, HCDR2 and HCDR3, wherein HCDR1, HCDR1 and HCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 100, 102 and 104, respectively;
- and
- (b) a LCVR comprising three light chain complementarity determining regions, LCDR1, LCDR2 and LCDR3, wherein LCDR1, LCDR2 and LCDR3 comprise the amino acid sequences of SEQ ID NOs:
- 84, 86 and 88, respectively, and
- wherein the refractory multiple myeloma is triple refractory to an anti-CD38 antibody, a proteasome inhibitor, and an immunomodulatory drug.
