Abstract
Provided is a functionalized polymer useful for extracting, isolating, and/or purifying membrane proteins with their native protein structures being retained. Also provided are a Native Cell Membrane Nanoparticle (NCMN) system which contains the above functionalized polymer and a membrane protein and a method to characterize and/assess the native structure of the membrane protein.
Claims26
A functionalized polymer obtained by
- (1) partially grafting a copolymer containing a plurality of acylating groups with an amine compound containing at least one primary or secondary amino group and two or more hydroxyl groups, and
- (2) treating the partially grafted copolymer with a basic aqueous solution to neutralize the remaining acylating groups,
- wherein the copolymer containing a plurality of acylating groups has a molecular weight of 5,000-18,000 Da and wherein the basic aqueous solution is NaOH aqueous solution, KOH aqueous solution, or ammonium aqueous solution.
