Abstract
Determination of the mechanism of allergy development in which a bacterium of the family Lachnospiraceae, which is an enteric bacterium, including Ruminococcus gnavus is involved and developing a method for preventing or treating allergies by intervening in the mechanism. Since the Ibp protein from a bacterium of the family Lachnospiraceae is involved in development of allergies, allergic reactions occurring in vivo can be prevented by administration or induction of in vivo production of a substance that binds to the Ibp protein.
Claims23
A pharmaceutical composition for treating or preventing an allergy, comprising a substance that binds to an Ibp protein or variant thereof of a bacterium of the family Lachnospiraceae.
A vaccine composition for preventing or treating an allergy, comprising a full-length or a portion of an Ibp protein or variant thereof of a bacterium of the family Lachnospiraceae as an immunogen.
A method for detecting the presence of a predisposition to developing an allergy in a subject, comprising:
- detecting an Ibp protein or variant thereof in a sample collected from a living subject;
- and
- detecting the Ibp protein or variant thereof in the subject, wherein the presence of the Ibp protein or variant thereof indicates that the subject has a predisposition to developing an allergy.
A method for detecting the presence of a predisposition to developing an allergy in a serum derived from a subject, comprising:
- (1) culturing human mast cells in the presence of the serum derived from a subject and measuring the amount of histamine in a culture supernatant;
- (2) adding and reacting IgG, IgM, or IgA to the serum from the subject, and then culturing human mast cells in the presence of the serum and measuring the amount of histamine in a culture supernatant;
- and
- (3) associating the amount of histamine obtained in (1) being higher than the amount of histamine obtained in (2) with the presence of a predisposition to developing an allergy in the blood of the subject.
A method for selecting an inhibitor against functional relations between IgE and an Ibp protein of a bacterium of the family Lachnospiraceae or variant thereof, comprising:
- (1) culturing mast cells in the presence of a test substance in the presence of IgE and an Ibp protein or variant thereof of a bacterium of the family Lachnospiraceae and measuring an amount of histamine in a culture supernatant;
- (2) culturing mast cells in absence of the test substance in the presence of IgE and the Ibp protein or variant thereof of a bacterium of the family Lachnospiraceae and measuring the amount of histamine in a culture supernatant;
- and
- (3) selecting the test substance as a substance having an activity of inhibiting functional relations between IgE and the Ibp protein or variant thereof of a bacterium of the family Lachnospiraceae, when the amount of histamine measured in (1) is lower than the amount of histamine measured in (2).
