Abstract
The present disclosure provides compounds of Formula I which can be tuned to provide agonistic or antagonistic toll-like receptor (TLR) bining properties. Further provided herein are pharmaceutical compositions comprising compounds described herein, as well as methods of using such compositions to treat a disease or condition in a subject.

A compound of Formula I:

- R1, R2, and R4 are each independently (C1-C6)alkyl, (C2-C6)alkenyl, (C2-C6)alkynyl, (C1-C6)alkanoyl, (C1-C6)alkoxycarbonyl, (C1-C6)alkanoyloxy, (C3-C6)-cycloalkyl, aryl, heteroaryl, or heterocycle, which can be optionally substituted with one or more groups independently selected from the group consisting of halo, cyano, hydroxy, oxo, oxiranyl, (C3-C5)cycloalkyl, optionally substituted aryl, and optionally substituted heteroaryl,
- or a pharmaceutically acceptable salt thereof, and
- wherein, when R4 is —C(═O)OCH3 and R2 is —CH2(p-Phe)—CH2NH2, R1 is not —CH2—CH2—CH2—CH3.
