Aspiration systems provided include first and second sheath assemblies, and a cannula. The first assembly includes a first shaft and hub. The first hub includes a hemostasis valve including a first multi-durometer sealing gland and a side port to be operatively coupled to a first suction source to generate a suction force. The cannula is configured to insert within the first assembly and be operatively coupled to a second suction source to generate another suction force. The first sealing gland is configured to selectively seal around the cannula or completely close when the cannula is not inserted. The second assembly includes a second shaft having a tapered distal end and second hub. The second hub includes a hemostasis valve having a second multi-durometer sealing gland. The first shaft is configured to insert into the second shaft, and the second sealing gland is configured to close around the first shaft.
