Abstract
The present application discloses a method of producing dopaminergic neurons from human stem cells by adding or increasing a concentration of vitamin into neurobasal media at approximately Day 20+/−3 of a protocol for differentiating pluripotent stem cells into dopaminergic neurons.
Claims36
A method of producing dopaminergic neurons from human stem cells comprising a step of adding or increasing the concentration of vitamin A to neurobasal media at approximately Day 20+/−3 of a protocol for differentiating pluripotent stem cells into dopaminergic neurons:
- wherein the produced dopaminergic neurons secrete greater than 30% more dopamine and its metabolites than dopaminergic neurons produced by the same differentiation protocol without the added or increased vitamin A.
A method of producing dopaminergic neurons from human stem cells comprising a step of adding or increasing the concentration of vitamin A to neurobasal media at approximately Day 20+/−3 of a protocol for differentiating pluripotent stem cells into dopaminergic neurons, in which the produced dopaminergic neurons are characterized by forming greater than 30% more neurites than dopaminergic neurons produced by a differentiation protocol without added or increased vitamin A.
A method of producing dopaminergic neurons comprising culturing naïve state stem cells in a neurobasal media, wherein the naïve state stem cells differentiate into dopaminergic neurons better than primed state stem cells.
