Abstract
The present disclosure provides methods and systems for generation of compositions comprising two or more sets of molecules. Compositions herein may comprise sets of molecules of different types. Sets of molecules may be generated by attachment to supports. Different types of molecules may be used to generate a large number of unique molecules. One or more types of molecules may be used to identify one or more additional types of molecules. Compositions of the present disclosure may be used in, for example, nucleic acid sequencing.

A method comprising:
- a) providing a partition among a plurality of partitions, wherein the partition comprises:
- i) a cell comprising a target nucleic acid, ii) a test agent or combination of test agents, and iii) a plurality of nucleic acid barcode molecules comprising a test agent-specific nucleic acid barcode sequence that identifies the test agent or combination of test agents, wherein the plurality of partitions comprises at least about 1,000 partitions;
- b) subjecting the partition to conditions sufficient to allow the test agent or combination of test agents to affect a cellular process identifiable by a change in an amount of the target nucleic acid in the cell;
- c) using the plurality of nucleic acid barcode molecules and molecules of the target nucleic acid or a derivative thereof to generate barcoded nucleic acid molecules, wherein the barcoded nucleic acid molecules comprise (i) the test agent-specific nucleic acid barcode sequence or a complement thereof, and (ii) a sequence of the target nucleic acid or a complement thereof.
