An engineered penicillin G acylase capable of acylating insulin, wherein the polypeptide sequence of said penicillin G acylase is at least 85% or more identical to SEQ ID NO:
- 40 and comprises a substitution as compared to SEQ ID NO:
- 40 at a position selected from positions 55, 71, 74, and 390.
