Abstract
An azide-containing cross-linker and method of preparing the same are provided. Such cross-linker can be used as an intermediate in a scalable production of a cross-linked hemoglobin protein having its beta subunits covalently linked to the azide-containing cross-linker. The present disclosure also pertains to a hemoglobin-based oxygen carrier (HBOC) comprising a pair of azide cross-linked hemoglobin proteins coupled by a copper (I) catalyzed azide-alkyne cycloaddition (CuAAC) and a method for preparing the same. The HBOC may be used in a method for increasing oxygen transport or for use in transfusions or perfusions.
Claims19
An azide-containing cross-linker, wherein the cross-linker is a compound of Formula I, II, III or IV
- wherein R is an alkyl;
- X is an alkali ion;
- n is 1 to 5;
- and
- R′ is a para- or meta-substitutable benzene or benzene isomer, a para- or meta-substitutable heterocyclic six-membered aromatic ring, a five membered aromatic ring, an indole ring, an indole derivative, a naphthalene or a naphthalene derivative, wherein each of the para- or meta-substitutable benzene, the para- or meta-substitutable benzene isomer, the para- or meta-substitutable heterocyclic six-membered aromatic ring, the five membered aromatic ring, the indole ring, the indole derivative, the naphthalene and the naphthalene derivative is optionally substituted.
