A chimeric antigen receptor (CAR), comprising:
- (a) an anti-Mucin 1* (MUC1*) antibody comprising:
- (i) a heavy chain (HC) variable region comprising a complementarity determining region 1 (HC-CDR1) comprising the amino acid sequence of SEQ ID NO:
- 123, a HC-CDR2 comprising the amino acid sequence of SEQ ID NO:
- 127, and a HC-CDR3 comprising the amino acid sequence of SEQ ID NO:
- 131;
- and
- (ii) a light chain (LC) variable region comprising a complementarity determining region 1 (LC-CDR1) comprising the amino acid sequence of SEQ ID NO:
- 173, a LC-CDR2 comprising the amino acid sequence of SEQ ID NO:
- 177, and a LC-CDR3 comprising the amino acid sequence of SEQ ID NO:
- 181;
- and
- (b) a cytoplasmic region comprising a CD3 zeta intracellular signaling domain comprising a mutation in a first immunoreceptor tyrosine-based activation motif (ITAM),
- wherein:
- when the CAR is expressed on a surface of an immune cell,
- the CAR binds specifically to a cancer cell, and
- the CAR has reduced signaling as compared to the same CAR but without the mutation in the first ITAM.
