Abstract
The present disclosure relates to a recombinant microorganism capable of efficiently producing L-leucine and an L-leucine production method using the same. The production method includes: (p) incubating a recombinant microorganism having an L-leucine biosynthesis pathway or a processed product of microbial cells thereof in a predetermined culture medium (X) to thereby produce L-leucine; and (q) recovering a fraction including the L-leucine from the predetermined culture medium (X). The recombinant microorganism includes, in an expressible form, a gene encoding an amino acid dehydrogenase. The amino acid dehydrogenase at least has a catalytic effect on an L-leucine production reaction in the L-leucine biosynthesis pathway.

A method for producing L-leucine, comprising:
- (p) incubating a recombinant L-leucine-producing microorganism or a processed product of microbial cells thereof in a predetermined culture medium (X) to thereby produce L-leucine;
- and
- (q) recovering a fraction including the L-leucine from a culture obtained in step (p),
- wherein the recombinant microorganism comprises in an expressible form a gene encoding an amino acid dehydrogenase, and the amino acid dehydrogenase has at least catalytic activity on an L-leucine production reaction.
A recombinant L-leucine-producing microorganism, comprising in a forcibly expressible manner:
- (i) a gene encoding a mutant type acetolactate synthase that has resistance to feedback inhibition of enzyme activity by at least one of valine, leucine and isoleucine;
- (iv) a gene encoding a mutant type 2-isopropylmalate synthase that has resistance to feedback inhibition of enzyme activity by L-leucine;
- and
- (ix) a gene encoding an amino acid dehydrogenase having at least catalytic activity on an L-leucine production reaction,
- wherein an ilvE gene or an ortholog thereof present on the genome has been inactivated or deficient in the recombinant microorganism.
