Abstract
The invention provides for systems, methods, and compositions for targeting nucleic acids. In particular, the invention provides non-naturally occurring or engineered RNA-targeting systems comprising a novel RNA-targeting CRISPR effector protein and at least one targeting nucleic acid component like a guide RNA.
Claims21
A non-naturally occurring or engineered composition for detecting and/or quantifying target RNA in a sample, said composition comprising:
- (a) a Cas 13a,
- (b) a guide molecule capable of forming a complex with the Cas13a and directing sequence-specific binding of the complex to a target sequence of a target RNA, and
- (c) an RNA-based masking construct comprising a non-target sequence,
- wherein the Cas13a exhibits collateral RNase activity and cleaves the non-target sequence of the RNA-based masking construct once activated by the target sequence.
A method of detecting and/or quantifying target RNA in a sample, said method comprising:
- (a) contacting a sample with a non-naturally occurring or engineered composition comprising:
- (i) a Cas13a,
- (ii) a guide molecule capable of forming a complex with the Cas13a and directing sequence-specific binding of the complex to a target sequence of a target RNA, and
- (iii) an RNA-based masking construct comprising a non-target sequence,
- wherein the Cas13a exhibits collateral RNase activity and cleaves the non-target sequence of the RNA-based masking construct once activated by the target sequence, and
- (b) detecting the target RNA based on the signal generated by cleavage of the RNA-based masking construct.
