Abstract
The present disclosure provides iPSC-derived microglia comprising a protective CD33 allele. Further provided herein are assays for screening analytes and genes associated with the protective CD33 allele.
Claims72
An isolated induced pluripotent stem cell (iPSC)-derived microglia cell line comprising a CD33 rs12459419T allele or CD33 rs12459419C allele.
A method for screening for a neurodegenerative disease comprising contacting an iPSC-derived microglia cell line comprising a CD33 rs12459419T allele with a sample.
The method of 26, wherein the pro-inflammatory (M1) agent is LPS, IFNγ, and/or GM-CSF.
A method of identifying a subject at risk for neurodegeneration comprising determining an expression level of at least 10 genes from Table 1A and at least 10 genes Table 1B in a blood sample, wherein a subject with decreased expression of genes in Table 1A and increased expression of genes in Table 1B as compared to a control is at risk for neurodegeneration.
A composition comprising a microglia cell population at least 90% positive for TREM2, CD45, CD11c, CD33, CD11b, PU.1, IBA-1, TREM2, CX3CR1, P2RY12, and/or TMEM119, wherein the microglia cell population is differentiated from iPSCs comprising a CD33 rs12459419T allele or CD33 rs12459419C allele.
The use of 63, wherein the pro-inflammatory (M1) agent is LPS, IFNγ, and/or GM-CSF.
