Abstract
The present invention provides antibodies and antibody-drug conjugates that bind to the extra domain B splice variant of fibronectin 1 and methods for preparing and using the same.
Claims26
1-44. (canceled)
A process for producing an antibody-drug conjugate,
- wherein the antibody-drug conjugate comprises:
- (a) an antibody, or antigen binding fragment thereof, that binds to extra domain B of fibronectin;
- (b) a linker;
- and
- (c) a drug;
- and
- wherein the process comprises at least one of the following:
- (a) linking the drug to the antibody, or antigen binding fragment thereof;
- (b) linking the linker to the drug;
- (c) linking the linker to the antibody, or antigen binding fragment thereof;
- (d) linking the antibody, or antigen binding fragment thereof to a conjugate of the linker and the drug.
A process for producing an antibody-drug conjugate,
- wherein the antibody-drug conjugate comprises:
- (a) an antibody, or antigen binding fragment thereof, that binds to extra domain B of fibronectin and that comprises a heavy chain comprising SEQ ID NO:25 and a light chain comprising SEQ ID NO:
- 31;
- (b) a cleavable linker;
- and
- (c) a cytotoxic agent;
- wherein the process comprises at least one of the following:
- (d) linking the drug to the antibody, or antigen binding fragment thereof;
- (e) linking the linker to the drug;
- (f) linking the linker to the antibody, or antigen binding fragment thereof;
- (g) linking the antibody, or antigen binding fragment thereof to a conjugate of the linker and the drug;
- and
- wherein the cleavable linker, the cytotoxic agent, and the antibody, or antigen binding fragment thereof, are linked to generate the structure:
- wherein:
- X—S represents the antibody, or antigen binding fragment thereof, and S is the sulfur atom of at least one cysteine residue in the antibody, or antigen binding fragment thereof.
A method of detecting extra domain B of fibronectin using a conjugate of an antibody, or antigen binding fragment thereof, that binds to extra domain B of fibronectin, wherein the antibody, or antigen binding fragment thereof, comprises at least one of the following:
- (a) a heavy chain variable region (VH) comprising the VH CDR1 region, VH CDR2 region, and VH CDR3 region of the VH amino acid sequence of SEQ ID NO:21;
- (b) a light chain variable region (VL) comprising the VL CDR1 region, VL CDR2 region, and VL CDR3 region of the VL amino acid sequence of SEQ ID NO:10.
