Abstract
Compositions useful for the detection of single molecules in a sample are provided. In some aspects, the disclosure provides a nucleic acid connected to a nucleotide and two or more luminescent labels. In some embodiments, the nucleic acids described herein comprise one or more structural features that provide enhanced fluorescence intensity. In some aspects, methods of sequencing using the labeled nucleotides of the disclosure are provided.
Claims26
A labeled molecule comprising a reactant connected to two or more luminescent labels via a linker, wherein each luminescent label is at least angstroms separated from any other luminescent label.

A labeled molecule comprising a reactant connected to two or more luminescent labels via a linker, wherein the linker is a nucleic acid comprising:
- a first oligonucleotide strand attached to two or more branching oligonucleotide strands at a terminal end of the first oligonucleotide via a covalent coupling compound, each branching oligonucleotide strand having a luminescent label;
- and
- a second oligonucleotide strand attached to a reactant, wherein the second oligonucleotide strand is hybridized with the first oligonucleotide strand.
A labeled molecule comprising a reactant connected to two or more luminescent labels via a linker, wherein the linker is a nucleic acid comprising:
- a first oligonucleotide component that comprises three or more oligonucleotide strands extending from a covalent coupling compound, wherein at least one of the oligonucleotide strands is attached to the reactant;
- and
- a second oligonucleotide component that comprises at least one oligonucleotide strand attached to a luminescent label, wherein the second oligonucleotide component is hybridized with the first oligonucleotide component, thereby connecting the reactant and the luminescent label.
