Abstract
Combining through self-modulation a flow of active media from a working vessel of a bioprocess together with a flow of reference media and making a time- and/or spatially-resolved referenced optical measurement of the active vs reference media in a confined flow region and time, such that the two liquids are measured in substantially identical conditions.

A system for monitoring a bioreactor comprising:
- a fluid handling system that produces an analysis media stream from active media extracted from the bioreactor, the active media containing at least one target media constituent from the bioreactor;
- a reference media source, wherein the reference media source is one of a fresh media source for feeding the bioreactor or a filter stage of the fluid handling system that filters the target media constituent from the active media extracted from the bioreactor;
- a fluid combiner that combines the analysis media with reference media from the reference media source into a flow chamber configured with an optical interrogation region;
- an optical characteristics measurement subsystem that measures optical characteristics of the analysis media and the reference media within the flow chamber;
- and
- an electronic processing system that calculates, based on the optical characteristics measurement, one or more of physical characteristics or biochemical characteristics of the at least one target media constituent.

A method of monitoring a bioreactor that produces a target media constituent, the method comprising:
- providing an analysis media stream from active media extracted from the bioreactor, the active media comprising the target media constituent;
- providing a reference media stream, wherein the reference media stream is provided from a fresh media source for feeding the bioreactor or from a filter stage that filters the target media constituent from the active media extracted from the bioreactor;
- combining the analysis media stream and the reference media stream into a flow chamber configured with an optical interrogation region;
- measuring a difference in optical characteristics between the analysis media stream and the reference media stream within the flow chamber;
- and
- calculating, with an electronic processing system, based on the optical characteristics measurement one or more of physical characteristics or biochemical characteristics of the target media constituent.

A method of monitoring an active media stream, the method comprising:
- receiving an analysis media that is derived from an active media that is derived from fresh media through bioreaction;
- combining the analysis media with a reference media into a flow chamber configured with an optical interrogation region;
- measuring optical characteristics of the analysis media and the reference media within the flow chamber;
- and
- calculating, with an electronic processing system, based on the optical characteristics measurement of the analysis media and/or the reference media, one or more of physical characteristics or biochemical characteristics of at least one target media constituent associated with the active media.
