Abstract
Provided herein is a reporter system for identifying a cytokine receptor modulator and uses thereof.
Claims25
A reporter system for identifying a cytokine receptor modulator, the system comprising:
- (a) a first fusion polypeptide comprising:
- (i) a first domain comprising a first subunit of a cytokine receptor;
- (ii) a second domain comprising at least a transcription activation domain and a DNA-binding domain of a transcription factor;
- and
- (iii) a linker linking the first and second domain, wherein the linker comprises a protease cleavage site between the first and second domain;
- and
- (b) a second fusion polypeptide comprising:
- (i) a first domain comprising a second subunit of the cytokine receptor;
- (ii) a second domain comprising at least a catalytic domain of a protease;
- and
- (iii) a linker linking the first and second domain;
- and
- (c) a reporter gene operably linked to a transcription factor regulatory element (TFRE),
- wherein the DNA-binding domain of a transcription factor recognizes and binds the TFRE.
