Abstract
This disclosure relates to methods of treating cancer using multi-specific binding proteins that bind NKG2D, CD16 and a tumor-associated antigen such as HER2, and pharmaceutical compositions thereof. Also provided are uses of the multi-specific binding protein in combination with an anti-PD-1 therapeutic and/or cytoskeletal disrupting chemotherapeutic agent.

1-102. (canceled)
A method of treating a non-small-cell lung cancer (NSCLC) in a subject in need thereof, the method comprising administering to the subject an effective amount of a multi-specific binding protein comprising:
- (a) a first antigen-binding site that binds NKG2D;
- (b) a second antigen-binding site that binds HER2;
- and
- (c) an antibody Fc domain or a portion thereof sufficient to bind CD16, or a third antigen-binding site that binds CD16,
- wherein the NSCLC has erbb2 gene amplification and/or an erbb2 activating mutation, and/or is a HER2-overexpressing cancer.
