Inhibitors of the MYST Family of Lysine Acetyl Transferases

A compound of Formula (I):

• wherein:
• R1 is phenyl optionally substituted with 1, 2, or 3 R1b, or 5- or 6-membered monocyclic heteroaryl optionally substituted with 1, 2, or 3 R1b;
• each R1b is independently selected from hydrogen, halo, C1-C6alkyl, C1-C6alkoxy, hydroxyalkyloxy, —O-alkylene-NR1b1R1b4, —O-alkylene-C(O)OR1b1, —O-alkylene-O-alkylene-NR1b1R1b4,
• cyano, —(CH2)0-2C(O)—OR1b1, —(CH2)0-2C(O)NR1b1R1b2, —(CH2)0-2NR1b1C(O)R1b3, —(CH2)0-2OH, and C3-C8-cycloalkyloxy;
• R1b1 is hydrogen or C1-C6alkyl;
• R1b2 is hydrogen or C1-C6alkyl;
• R1b3 is hydrogen or C1-C6alkyl;
• and R1b4 is hydrogen,
• R2 is:
• one X1 is CR3 and the other X1 are independently selected from CR2b;
• each R2b is independently hydrogen, halo, —(CH2)0-2OH, C1-C3alkyl, cyclopropyl, cyano, —CHF2, —CF3, C1-C4alkoxy, —OCHF2, —OCF3, or C3-C8cycloalkyloxy;
• R2d is hydrogen, halo, C1-C6alkyl, C1-C6cycloalkyl, C1-C6alkoxy, or C3-C8-cycloalkyloxy;
• R3 is —(CH2)0-2-L-Y;
• L is a bond or O;
• Y is a 5-membered monocyclic heteroaryl substituted with RY and optionally substituted with R2e;
• Y is a 6-membered monocyclic aryl or heteroaryl substituted with RY and optionally substituted with 1 or 2 R2e;
• or Y is a 8- or 9-membered bicyclic heterocyclic substituted with RY and optionally substituted with 1 or 2 R2e;
• each R2e is independently hydrogen, —OH, halo, C1-C6alkyl, halo-C1-C6alkyl, C3-C8cycloalkyl, C1-C6alkoxy, halo-C1-C6alkoxy, or C3-C8cycloalkyloxy;
• RY is —(CH2)0-3NR3bC(O)R3a, —(CH2)0-2NR3bS(O)2R3a, —C(O)R3a, —S(O)2R3a, —C(O)NR3bR3a, or C3-C8heterocycloalkyl substituted with —C(O)R3a
• R3a is selected from C1-C6alkyl substituted with 1 or 2 halo which are independently selected;
• C1-C6alkyl substituted with cyano;
• C1-C6alkyl substituted with fluoroalkoxy;
• C1-C6alkyl substituted with aryloxy or heteroaryloxy, each of which is optionally substituted with 1-3 substituents each independently selected from halo, C1-6alkyl, C2-6alkenyl, C1-6alkoxy, cyano, C3-8cycloalkyl or C3-8heterocycloalkyl;
• C2-C6alkenyl;
• C2-C6alkenyl substituted with cyano;
• C2-C6alkenyl substituted with halo;
• —CH═CH—CH2—NR3cR3d;
• —CH═CH—CH2—O—C1-C6alkyl;
• C3-C8cycloalkenyl;
• —C(O)—C3-C8cycloalkyl;
• C2-C6alkynyl;
• —CH≡CH—CH2—NR3cR3d;
• CH≡CH—CH2—OH;
• —CH≡CH—CH2—O—C1-C6alkyl;
• spirocycloalkyl substituted with cyano;
• chloropyridyl;
• fluoropyridyl;
• chloropyrazinyl;
• fluoropyrazinyl;
• chloropyrimidinyl;
• fluoropyrimidinyl;
• pentafluorophenyl;
• tetraflurophenyl;
• trifluorophenyl, difluorophenyl;
• and monofluorophenyl;
• R3b is selected from hydrogen, and C1-C6alkyl;
• or a pharmaceutically acceptable salt or salts thereof;
• and/or a stereoisomer or mixture of stereoisomers thereof.
